Gram Negative Strips

Rapid diagnostic test for aminopeptidase enzyme present in the cell membrane of Gram negative bacteria.

Gram Negative Strips provide rapid differentiation of Gram–positive and Gram-negative aerobes and facultative anaerobes. The test detects cell wall aminopeptidase present in Gram-negative organisms by using the chromogenic substrate L-aniline-4-nitroanilide (LANA). Each strip is impregnated with LANA (colourless), and the aminopeptidase hydrolyses LANA yielding bright yellow nitroaniline. Gram-positive organisms do not produce any colour change. The technique is convenient, reliable and clean! It has been shown to work in the presence of non-cultivable organisms, allowing their Gram classification to be assigned.

Description: Paper strip impregnated with L-alanine-4-nitroanilide.

Method of use: First moisten printed zones on strip with activating agent from dropper bottle. Using wire or plastic loop (such as Microloop), touch 2 or 3 colonies of test organism on agar medium. Smear organism onto test zone on the strip. Positive result is indicated within a one minute by smeared area turning bright yellow.
