Chromic acid oxidation forms aldehydes from fungal cell wall polysaccharide components, which are subsequently demonstrated by reduction of an alkaline hexamine-silver complex. The reaction may be compared to that of the periodic acid Schiff reaction.

Grocott's alkaline hexamine-silver solution allows silver ions upon reduction, to appear dark or black, also known as "argentaffin reaction" the ability of a silver complex solution to blacken a tissue element without the need of a reducing bath.

Kit Components:
3.8% chromium trioxide aqueous solution (500 ml)
1% sodium bisulfite aqueous solution (500 ml)
5% silver nitrate aqueous solution (45 ml)
Methenamine solution (900 ml)
5% Borax aqueous solution (250 ml)
0.2% gold chloride aqueous solution (500 ml)
2% Sodium thiosulfite aqueous solution (500 ml)
0.5% Light Green stock solution (100 ml)
