HOPE® = Hepes-glutamic acid buffer mediated Organic solvent Protection Effect. Hope is a combination of amino acids and glucose blended together with an organic buffer. Dehydration by acetone as the single dehydrating agent and pure paraffin denature proteins and preserve nucleic acids and antigenic-structures allowing clearly visible molecular level results.

New fixation-technique for cells and tissues utilizing the revolutionary HOPE®-fixative. Scientists have developed an exciting new fixative, which has started to greatly enhance the possibilities of tissue paraffin-embedded archival materials.

The HOPE® technique is mainly used for the preparation of paraffin embedded tissue sections. In contrast to other fixation methods HOPE® does not completely denature or cross-link structural proteins, enzymes, and nucleic acids.

Applications for immunohistochemistry, in situ hybridization targeting RNA and DNA, PCR and RT-PCR have been demonstrated using properly prepared paraffin embedded tissue sections. Tissue specimens fixed with the HOPE technique allow greater morphology and retrospective studies on the molecular level.

The HOPE®-Fixation technique requires HOPE® Fixative System I and HOPE® Fixative System II to complete the protocol.
